Tigecycline
Indications/Uses
Complicated skin & skin structure infections & intra-abdominal infections in patients ≥18 yr.
Dosage/Direction for Use
Initially 100 mg, followed by 50 mg every 12 hr by IV infusion over approx 30-60 min for 5-14 days. Hepatic impairment 100 mg followed by 25 mg every 12 hr.
Contraindications
Special Precautions
Known hypersensitivity to tetracyclines. Discontinue use if Clostridium difficile-associated diarrhoea is suspected or confirmed. Pseudomembranous colitis; pancreatitis; permanent tooth discoloration during tooth development; photosensitivity, pseudotumor cerebri, anti-anabolic action. Superinfection or overgrowth of non-susceptible organisms including fungi. Severe hepatic impairment. Not to be used during pregnancy. Lactation. Not recommended in childn <18 yr.
Adverse Reactions
Nausea, vomiting, diarrhea. Sepsis/septic shock, pneumonia, abscess, infections; prolonged activated partial thromboplastin & prothrombin time; hypoglycaemia, hypoproteinaemia; dizziness; phlebitis; abdominal pain, dyspepsia, anorexia; elevated serum AST & ALT, hyperbilirubinaemia; pruritus, rash; impaired healing, inj site reaction, headache; elevated serum amylase, increased BUN.
Drug Interactions
Decreased clearance & increased AUC w/ warfarin. Reduced effectivity of OCs. Incompatibilities w/ amphotericin B, amphotericin B lipid complex, diazepam, esomeprazole, omeprazole.
